Effect of puerarin on cerebral ischemia-reperfusion injury in rats
摘要目的探讨葛根素对大鼠脑缺血-再灌注(I-R)损伤的影响及其机制。方法将72只SD大鼠随机均分为假手术组、I-R组和葛根素组。I-R组和葛根素组采用大脑中动脉梗阻法造模,缺血2 h后予再灌注;葛根素组在造模前3 d及造模后予肌肉注射葛根素注射液30 mg/kg。再灌注3、6、12和24 h时对各组大鼠行神经功能缺损评分、磁共振扩散峰度成像检查和神经元细胞HE染色,ELISA法检测脑组织炎症因子水平。再灌注24 h时采用Western blot检测脑组织蛋白酪氨酸激酶(JAK)/信号转导及转录激活因子(STAT)信号通路蛋白的表达。另取6只SD大鼠作为空白对照(空白组)。结果与假手术组比较,各时间点I-R组神经功能缺损评分、平均扩散峰度值(MK)、TNF-α、IL-6、IL-8水平和JAK1、JAK3、STAT1、STAT3蛋白表达水平升高,平均扩散系数(MD)降低(P<0.05)。与I-R组比较,各时间点葛根素组神经功能缺损评分、MD、MK、TNF-α、IL-6、IL-8水平和JAK1、JAK3、STAT1、STAT3蛋白表达水平均降低(P<0.05)。HE染色结果显示,空白组和假手术组神经元细胞形态正常,I-R组各时间点神经元细胞坏死和凋亡明显增多,葛根素组神经元细胞病理损伤较I-R组减轻。结论葛根素通过抑制JAK/STAT信号通路激活,减轻下游炎症因子释放,从而发挥脑保护作用。Objective To explore the effect of puerarin on cerebral ischemia-reperfusion(I-R) injury and its mechanism in rats.Methods Seventy-two SD rats were equally and randomly divided into three groups.Group A accepted sham operation.The cerebral I-R injury model was established by middle cerebral artery obstruction for 2 hours,which was followed by reperfusion in groups of B and C.Group C accepted intramuscular injection of puerarin 30 mg/kg at 3 days before and after modeling.At 3,6,12 and 24 hours after reperfusion,neurological deficit score,magnetic resonance diffusion kurtosis imaging and neuron HE staining were performed and the levels of inflammatory factors in brain tissues were detected by ELISA.The protein expressions of Janus kinase(JAK)/signal transducer and activator of transcription(STAT) signaling pathway was detected by Western blot at 24 hours after reperfusion.Another 6 SD rats were taken as the blank controls(group D).ResultsCompared with group A,neurological deficit score,mean kurtosis(MK),levels of TNF-α,IL-6,IL-8 and protein expression dwrtosis of JAK1,JAK3,STAT1 and STAT3 in brain tissues were increased,while mean diffusivity(MD) was decreased in group B at each time point(P<0.05).Compared with group B,neurological deficit score,MD,MK,levels of TNF-α,IL-6,IL-8 and protein expressions of JAK1,JAK3,STAT1 and STAT3 in brain tissues were decreased in group C at each time point(P<0.05).The results of HE staining showed that the morphology of neurons in groups of A and D was normal,the necrosis and apoptosis of neurons in group B were increased,and the pathological injury of neurons in group C was less than that in group B at each time point.Conclusion Puerarin plays a protective role in the brain by inhibiting activation of JAK/STAT signaling pathway and reducing the release of downstream inflammatory factors.
