目的:观察小青龙汤对小青龙汤证模型大鼠肺组织中磷脂酰肌醇3激酶(PI3K)、蛋白激酶B(Akt)、哺乳动物雷帕霉素靶蛋白(mTOR)mRNA表达的影响。方法:将24只雄性Wistar大鼠按照完全随机分组分成空白组、模型组、小青龙汤高剂量组、小青龙汤低剂量组共4组。建立外寒里饮(小青龙汤证)大鼠模型,予小青龙汤灌胃治疗15 d后,记录各组大鼠的体质量变化并加以分析,运用实时荧光定量(RT-qPCR)技术检测各组大鼠肺组织中PI3K、Akt、mTOR mRNA表达的差异。结果:与空白组比较,模型组与小青龙汤高、低剂量组大鼠体质量在造模后增加不明显,甚至出现负增长(P<0.01);经过药物干预治疗后,与空白组比较,模型组大鼠体质量增长仍不明显;与模型组比较,小青龙汤高、低剂量组大鼠体质量明显增加(P<0.01)。RT-qPCR结果显示,与空白组比较,模型组PI3K、Akt、mTOR mRNA表达升高(P<0.05);与模型组比较,小青龙汤高剂量组PI3K、Akt、mTOR mRNA表达降低(P<0.05,P<0.01),小青龙汤低剂量组PI3K、Akt、mTOR mRNA表达有降低趋势,但差异无统计学意义。结论:小青龙汤证的发生发展与PI3K、Akt、mTOR mRNA表达量的上调相关,小青龙汤高剂量治疗效果明显,其作用机制与其能够下调PI3K、Akt、mTOR mRNA的相对表达量有关。Objective:To observe the effects of the Xiaoqinglong decoction(XQLD)on the relative expressions of PI3K(phosphatidylinositol-3 kinase),Akt(protein kinase B),mTOR(mammalian rapamycin target protein)mRNA in the lung tissue of XQLD syndrome model rats.Methods:24 male Wistar rats were randomly divided into 4 groups:normal group,model group,high-dose XQLD group and low-dose XQLD group.XQLD syndrome models were established,then rats in treatment groups were gave XQLD for 15 days.The changes of weight of rats were recorded and expressions of PI3K,Akt,mTOR mRNA were detected by Quantitative Real-time PCR(RT-qPCR).Results:Compared with the model group,the weight of rats was increased in XQLD group(P<0.01).Compared with the blank group,the expressions of PI3K,Akt,and mTOR mRNA in the model group were increased(P<0.05);compared with the model group,the expressions of PI3K,Akt,and mTOR mRNA in the high-dose XQLD group were decreased significantly(P<0.05 or P<0.01).Conclusion:The occurrence and development of XQLD syndrome is related to the up-regulation of expressions of PI3K,Akt and mTOR mRNA.The therapeutic mechanism of XQLD may be related to its ability to down-regulate the relative expressions of PI3K,Akt and mTOR mRNA.
