舟山海域常见经济物种组氨酸二肽的提取和鉴定

摘要 组氨酸二肽是一类广泛存在于生物体内的活性肽,具有多种重要的生理学活性,在食品、医药等领域具有重要应用价值。为了解舟山海域常见经济水产类物种的组氨酸二肽含量,采取邻苯二甲醛显色法和液相色谱-质谱联用法,对舟山海域23种硬骨鱼类,6种甲壳类和6种软体动物,同时,选择金枪鱼、鲣鱼、三文鱼和鸡胸肉作为参照,共计39个物种,开展其肌肉组织中组氨酸二肽的含量测定。结果显示,采用邻苯二甲醛显色法,在上述所有物种中均能检测到组氨酸二肽;进一步采用液相色谱-质谱联用法,分析上述物种中,肌肽,鹅肌肽和高肌肽三种组氨酸二肽成员的含量,结果表明鱼类肌肉组织中的组氨酸二肽以鹅肌肽为主;而在甲壳类生物以及软体动物类中则基本未鉴定到组氨酸二肽。进一步的抗氧化活性测试结果表明,银鲳、银姑鱼和厚壳贻贝的组氨酸二肽提取物具有较高的DPPH清除率。上述研究为了解舟山海域水产物种的营养价值以及海洋生物组氨酸二肽资源的开发奠定了基础。 Histidine-containing dipeptide is a family of non-protein nitrogen-containing compound with multiple physiological roles mainly present in excitable tissues of vertebrates and with valuable applications in the fields of food and medicine.For exploring the distribution of histidine-containing dipeptides in economic aquatic species collected from Zhoushan fishing ground of East China Sea,the concentration of histidine-containing dipeptides in 23 bony fishes,6 crustaceans,and 6 mollusks were detected using o-phthalic aldehyde chromogenic method and liquid chromatography-mass spectrum,respectively.In addition,the tuna,bonito,salmon,and chicken breast were selected as reference samples.The results showed that histidine-containing dipeptides can be detected in all species by o-phthalic aldehyde chromogenic method.Furthermore,the contents of three histidine dipeptides,carnosine,anserine,and homocarnosine,were analyzed by liquid chromatography-mass spectrometry.The anserine is the main component of histidine-containing dipeptides in fish muscle.However,no histidine-containing dipeptide can be detected in crustaceans and molluscs by the method of chromatography-mass spectrum.Further antioxidant activity tests showed that the extracts of histidine-containing dipeptides from Pampus argenteus,Pennahia argentata,and Mytilus coruscus have higher DPPH clearance.The present findings greatly expanded the data of histidine-containing dipeptide distribution and provided a clue for understanding the roles of histidine-containing dipeptide in different animals,and also nutritional assessment for marine aquatic species.
