不同保存条件下龙头鱼不同组织DNA提取效果比较分析

摘要 为探索龙头鱼分子标记实验的基因组DNA的最佳提取组织和保存方法,本实验以肌肉和鳍条组织为研究对象,对短期冷藏保存(4℃)、冷冻保存(-20℃)、无水乙醇保存、75%乙醇保存和10%甲醛保存这五种状态下的样本组织进行DNA提取,并用紫外分光光度计、基因组DNA琼脂糖凝胶电泳和PCR扩增产物琼脂糖凝胶电泳检测所提取DNA的质量。实验结果表明,DNA浓度肌肉组织高于鳍条组织,10%甲醛保存DNA浓度明显低于其他保存方法;除10%甲醛保存样品外,其他各保存方法及不同组织提取的DNA质量均可满足PCR扩增。 In order to explore the best tissue and preserving method for genomic DNA extraction for the molecular marker experiment of Harpodon nehereus,the experiment took muscle and fin tissues as the research objects.These tissues were preserved under different conditions:refrigerated for short term(4℃),frozen(-20℃),fixed in anhydrous ethyl alcohol,75%alcohol and 10%formaldehyde.The quality of DNA was detected by ultraviolet spectrophotometer,agarose gel electrophoresis and PCR amplification.The results showed that the concentration of DNA in muscle tissue is higher than that in fin tissue,and the concentration of DNA fixed in 10%formaldehyde is significantly lower than other preservation methods.Except the 10%formaldehyde preserved samples,the quality of DNA extracted from different tissues and preservation methods could meet the requirements of PCR amplification.
