摘要 采用L9(34)正交实验法,优化了微波辅助提取刺松藻中多酚的工艺条件,并对其抗氧化活性、结构等进行了探究,得出最佳提取条件为液固比为1:25,微波辐射时间为55 s,微波辐射功率500 W,多酚含量为2.76 mg·g-1。由光谱分析推测提取物中可能含有黄烷醇类、黄烷酮类等物质,结构中可能含有νO-H、νC-H、νC-O、γ=C-H等振动;以Vc为阳性对照,探究多芬提取物的抗氧化活性。结果表明,刺松藻多酚DPPH自由基、羟自由基半抑制质量浓度IC50分别为20.12μg·mL-1、12.56μg·mL-1,浓度为10μg·mL-1时,对Fe2+诱导卵黄脂蛋白脂质过氧化抑制率为6.74%,为其进一步开发利用奠定了基础。 In this experiment,L9(34)orthogonal test was used to optimize the microwave-assisted extraction process of polyphenols from Codium fragile Hariot.,and its antioxidant activity and structure analysis were studied.The results showed that the optimum extraction conditions were liquid-solid ratio of 1:25,microwave radiation time of 55 s,microwave radiation power of 500 W,the polyphenols content could achieve up to 2.76 mg·g-1.The spectral analysis suggested that the extract might contain flavanols,flavanones and other substances,and the structure might contain vibration such asνO-H,νC-H,νC-O,γ=C-H,etc.Compared with Vc,the antioxidant activity of the extract was investigated.The IC50 of DPPH radical and hydroxyl radical is 20.12μg·mL-1 and 12.56μg·mL-1,respectively.At the concentration of 10μg·mL-1,the lipid peroxidation of vitellin induced by Fe2+was inhibited andtheinhibition rate is 6.74%.The results in this study lay a foundation for its further development and utilization.
