摘要 目的:探讨雷帕霉素对上颌窦黏膜细胞成骨分化的影响。方法:上颌窦黏膜细胞经不同浓度的自噬激动剂雷帕霉素处理后诱导成骨分化,然后进行碱性磷酸酶活性检测、茜素红染色以及自噬基因和成骨基因转录水平的检测,评估上颌窦黏膜细胞成骨分化和自噬水平的改变。结果:碱性磷酸酶活性、钙结节形成量以及成骨基因的表达随成骨诱导逐渐增加,并且100 nmol/L的雷帕霉素处理组表现出较强的成骨能力和自噬水平。结论:合适浓度的雷帕霉素可促进上颌窦黏膜细胞的成骨分化,这一过程可能与自噬途径相关。 Objective:To investigate the effects of rapamycin(RAPA)on the osteogenic differentiation of maxillary sinus membrane cells(MSMCs).Methods:MSMCs were treated with different concentrations of RAPA and induced to differentiate into osteocytes.Alkaline phosphatase(ALP)activity,alizarin red staining and the transcriptional levels of autophagic and osteogenic marker genes were detected to evaluate the level of osteogenic differentiation and autophagy of MSMCs.Results:After RAPA treatment and osteogenic induction,ALP activity,calcium nodules formation,and the expression levels of osteogenic marker genes of MSMCs increased.Superior expression levels of osteogenesis and autophagy occurred in 100 nmol/L RAPA treated MSMCs.Conclusion:Approximate concentration of RAPA can promote osteogenic differentiation of MSMSCs,indicating a possible relationship between osteogenic differentiation and autophagy of MSMCs.
