摘要 【目的】筛选番茄抗南方根结线虫侵染相关基因,为其克隆及番茄抗病育种提供参考依据。【方法】用Illumina HiSeqTM4000对南方根结线虫侵染和未侵染番茄抗性材料F5样本的转录组进行测序,基于差异表达基因本位数据库(GO)和Pathway显著性富集(KEGG)分析,挖掘参与抗南方根结线虫的相关基因。【结果】筛选到1116个差异表达基因,其中731个为上调基因,385个为下调基因,其功能归类于生物过程、细胞组分和分子功能三大类1830个条目,富集于94条KEGG代谢通路。差异表达基因的GO和KEGG分析结果表明,植物与病原互作相关的22个基因差异表达,其中18个抗性相关基因上调表达,上调表达明显的抗性基因包括12个Ca+-CaM/CML信号相关基因(11个类钙调蛋白、1个钙调蛋白)、3个WRKY转录因子和1个环核甘酸门控离子通道蛋白。【结论】Ca+-CaM/CML信号、WRKY转录因子和环核甘酸门控离子通道是番茄抗性材料F5对南方根结线虫抗性反应的重要机制,可作为深入探索根结线虫与番茄互作及挖掘番茄关键抗性相关基因的参考依据。 【Objective】The resistance genes related to infection response of tomato and nematode were screened to provide the basis for mining the key resistance related genes and tomato disease resistance breeding.【Method】Illumina HiSeqTM4000 technique was used to sequence the tomato resistant material F5 genes expression level infected and not infected by Meloidogyne incognita.On the basis of GO and KEGG analysis of the differentially expressed genes,genes resistant to M.incognita were mined.【Result】1116 differentially expressed genes were obtained,among which 731 genes were up-regulated,and 385 genes were down-regulated.The functions of these differentially expressed genes were classified into 1830 GO items in three categories:biological process,cell components and molecular functions,and were enriched in 94 KEGG metabolic pathways.GO database and KEGG metabolic analysis on differentially expressed genes revealed 22 genes related to plant-pathogen interaction were differentially expressed,among which 18 genes were up-regulated.The significantly up-regulated resistant genes included 12 Ca+-CAM/CML signal related genes(11 calmodulin-like protein CML,1 calmodulin),3 WRKY transcription factor,a protein CNGC.【Conclusion】Ca+-CAM/CML signal,WRKY transcription factor and cyclic glycolate gated ion channel are important mechanisms of F5 resistance to M.incognita.It can provide reference for exploring interaction between meloidogyne and tomato and mining tomato key resistance related genes.
