黑龙江省种植大豆品种遗传多样性分析及与性状关联SSR标记筛选

摘要 为从分子水平鉴定黑龙江省种植大豆品种的遗传多样性,并筛选与蛋白质、脂肪、百粒重和水分等大豆主要性状显著关联的SSR分子标记,利用18对SSR引物,采用荧光毛细管电泳法分析黑龙江省75个种植大豆品种遗传多样性,并与性状进行关联分析。结果表明:18个SSR标记中的10个标记可以实现单对引物准确鉴别42个大豆品种,18个SSR标记的等位变异的数目变化为3(Satt387)~12个(Satt100),平均每个位点等位变异数为7.333个,多态信息指数范围为0.146(Sat_084)~0.675(Satt442),平均多态信息指数为0.405;群体结构分析发现,该群体分为3个亚群;性状关联分析表明,标记Satt175和Satt100与脂肪存在显著关联,标记Satt514和Satt294与百粒重存在显著关联,其中Satt294与百粒重和水分均存在显著关联。研究结果为探索大豆品种间的亲缘关系,挖掘大豆相关性状的关联位点,验证大豆种质资源的真实性,以及保障黑龙江省大豆品牌和食品安全提供参考。 In order to identify the genetic diversity of soybean varieties grown in Heilongjiang Province at the molecular level,and to screen SSR molecular markers that are significantly related to the main traits of soybeans,such as protein,fat,100-seed weight,and moisture,the genetic diversity of 75 soybean varieties planted in Heilongjiang province and the correlation with the traits were analyzed in this study,using 18 pairs of SSR primers and fluorescent capillary electrophoresis.Results indicated that 10 among the 18 SSR markers can accurately identify 42 soybean varieties with a single pair of primers.The number of allelic variations of the 18 SSR markers varied from 3(Satt387)to 12(Satt100)on the average of 7.333.The polymorphic information content(PIC)among genotypes was at the range of 0.146(Sat_084)to 0.675(Satt442)with an average of 0.405.Group structure analysis indicated that the group could be divided into 3 subgroups.Through trait association analysis,it could be found that that both Satt175 and Satt100 markers were significantly associated with fat,markers Satt514 and Satt294 were associated with 100-seed weight,and Satt294 was also significantly associated with 100-seed weight and moisture,respectively.Therefore the present results of this study provide reference for exploring the genetic relationship among soybean varieties,digging the related sites of soybean-related traits,verifying the authenticity of soybean germplasm resources as well as brand protection and quality safety in Heilongjiang Province.
