《缺氧环境对鼻咽纤维血管瘤细胞增殖、侵袭的影响及可能的作用机制》为作者:陆徐最新的研究成果,本论文的主要观点为目的:探讨缺氧环境下培养鼻咽血管纤维瘤(nasopharyngeal angiofibroma,NA)细胞的生物学功能变化以及可能的分子机制。方法:将NA细胞置于不同浓度的氧气环境(20%、1%O2)中培养,CCK-8法检测NA细胞的增殖活性,Transwell检测NA细胞的侵袭能力,QPCR或western blot检测NA细胞中HIF-1α、VEGF、TGF-β、ERα的表达。结果:与常氧状态相比,缺氧状态下的NA细胞增殖活性增强(P<0。001),侵袭能力显著上升(P<0。001),HIF-1α、VEGF、TGF-β表达均上调,ERα表达下调(P均<0。05)。结论:缺氧环境下,NA细胞可能通过上调HIF-1α、VEGF、TGF-β表达,下调ERα表达,来增强细胞的增殖、侵袭能力,促进NA进展。 关键词:缺氧;鼻炎纤维血管瘤;HIF-1α Abstract: Objective: To investigate the changes of biological function and possible molecular mechanism of nasopharyngeal angiofibroma (NA) cells cultured in hypoxia environment。 Methods: NA cells were cultured in different concentrations of oxygen (20%, 1% O2)。 The proliferative activity of NA cells was detected by CCK-8 method, the invasiveness of NA cells was detected by Transwell, and the expression of HIF-1 α, VEGF, TGF-β, ERα in NA cells was detected by qPCR or Western blot。 Results: compared with the normoxia state, the proliferation activity of NA cells in hypoxia state was enhanced (P < 0。001), the invasion ability was significantly increased (P < 0。001), the expression of HIF-1 α, VEGF, TGF-β was up-regulated, and the expression of ERα was down regulated (P<0。05)。 Conclusion: under hypoxia environment, Na cells may enhance the proliferation and invasion of cells and promote the progress of Na by up regulating the expression of HIF-1α, VEGF, TGF-β and down regulating the expression of ERα。 Keywords: hypoxia; rhinitis fibroangioma; HIF-1α。不知是否符合录用要求,望您批评与指正。
