基于网络药理学研究斑蝥复方制剂治疗前列腺癌的作用机制

摘要 目的基于网络药理学探索斑蝥复方制剂治疗前列腺癌(PCa)的潜在活性成分以及作用机制。方法通过TCMSP数据库筛选斑蝥复方制剂的活性成分及对应靶点蛋白,利用GeneCards和OMIM数据库平台筛选出PCa的相关靶点,应用Cytoscape 3.7.2构建网络图,以STRING在线分析平台构建靶蛋白相互作用PPI网络并进行分析,并在GEPIA数据库中验证关键靶点基因在PCa组织中的表达,并运用DAVID数据库对靶点进行基因本体论(GO)生物过程分析以及京都基因与基因组百科全书(KEGG)信号通路富集分析。结果筛选出斑蝥复方制剂的42个有效活性成分和240个有效靶点,药物-疾病共同靶点120个。PPI网络发现TP53、AKT1、MYC、JUN、CASP3、TNF、MMP9、PTEN等可能是斑蝥复方制剂治疗PCa的关键靶点。GO功能分析发现254个条目,涉及转录因子活性、蛋白质活性、DNA活性、ATP活性、细胞因子活性等以及生物合成过程的调控等方面。KEGG通路分析发现82个条目,涉及癌症通路、PI3K-Akt信号通路、癌症蛋白聚糖通路、癌症微小RNA通路、丝裂原活化蛋白激酶信号通路、肿瘤坏死因子信号通路等信号通路。结论本研究通过网络药理学方法预测斑蝥复方制剂治疗PCa是多成分、多靶点、多通路的调节,这为今后的分子生物学研究和临床应用提供了一定参考。 Objective To explore the potential active ingredients and mechanism of action of cantharidin compound preparations in the treatment of prostate cancer based on network pharmacology.Methods Screen the active ingredients and corresponding target proteins of cantharidin compound preparations through the TCMSP database,screen out the relevant targets of prostate cancer using GeneCards and OMIM database platforms,use Cytoscape 3.7.2 to construct a network diagram,and use the STRING online analysis platform to construct target proteins Interact with the PPI network and analyze it,verified the expression of key target genes in prostate cancer tissues in the GEPIA database,and use the DAVID database to analyze the GO function and KEGG pathway enrichment of the target.Results Forty-two effective active ingredients and 240 effective targets of cantharidin compound preparations and 120 common drug-disease targets were screened out.PPI network found that TP53,AKT1,MYC,JUN,CASP3,TNF,MMP9,PTEN,etc.may be the key targets of cantharidin compound preparations for the treatment of prostate cancer.GO function analysis revealed 254 items,involving transcription factor activity,protein activity,DNA activity,ATP activity,cytokine activity,etc.,as well as the regulation of biosynthetic processes.KEGG pathway analysis revealed 82 entries,involving cancer pathways,PI3K-Akt signaling pathways,cancer proteoglycan pathways,cancer microRNA pathways,MAPK signaling pathways,tumor necrosis factor signaling pathways and other signaling pathways.Conclusions This study analyzed and predicted the molecular mechanism of cantharidin compound preparations in the treatment of prostate cancer through network pharmacology,and found that its effect was through multi-component,multi-target,and multi-path regulation,which provides reference for future molecular biology research and clinical applications.
