摘要 本实验旨在探究核糖体蛋白基因(RPL35A)在不同月龄寒泊羊肌肉组织中的mRNA表达及其生物学功能。选取1月龄、7月龄和13月龄寒泊公羊各3只,采集背最长肌,利用转录组测序(RNA-seq)与荧光定量PCR(qRT-PCR)技术相结合,分析比较RPL35A基因的表达丰度。应用生物信息学方法分析RPL35A基因的核苷酸和氨基酸序列的分子结构特征、蛋白质特性及网络互作。结果表明:1月龄寒泊羊背最长肌RPL35A基因mRNA高于7月龄,后者低于13月龄,RNA-seq与qRT-PCR结果趋势一致。绵羊RPL35A基因核苷酸及氨基酸序列与山羊、牛、家马和猪的相似性较高;RPL35A编码蛋白为碱性蛋白(PI=11.07)、亲水蛋白(亲水值为-0.579),稳定性较好,属于非跨膜蛋白,在线粒体(69.6%)、细胞核(17.4%)、细胞质(8.7%)、过氧化物酶体(4.3%)内发挥作用。蛋白互作网络分析表明RPL35A蛋白在核糖体合成过程中发挥着重要作用。综上,RPL35A基因可能参与寒泊羊肌肉生长发育过程。 The aim of this study was to analyze the biological function and expression level of ribosomal protein L35 A(RPL35A)of different months age muscle tissues from Hanper sheep.Longissimus lumborum were collected from three rams of 1-month-age,7-month-old and 13-month-old Hanper sheep,respectively.The expression abundance of RPL35A gene was studied and compared by RNA-seq and qRT-PCR.The molecular construction,proteins characteristics and network interactions of the nucleotide and amino acid sequences of RPL35A gene were analyzed by bioinformatics.The results showed that the mRNA expression of RPL35A gene in 1-month-age was greater than that of 7-month-age,the latter was lower than that of 13-month-age.The result trend of RNA-seq was consistent with that of qRT-PCR.The nucleotide and amino acid sequences of ovine RPL35A gene was similar with goats,cattle,horses and pigs.The protein encoded by RPL35A,belonging to non-transmembrane protein,is alkaline(PI=11.07),hydrophilic(hydrophilic value is-0.579),with better stability,which play a role at mitochondrion(69.6%),nucleus(17.4%),cytoplasm(8.7%),and peroxisomes(4.3%),separately.It play an important function of RPL35A protein in the synthesis of ribosomes through protein interaction network analysis.In conclusion,the RPL35 A gene may participate in process of growth and development of muscle tissue in Hanper sheep.
