摘要 目的研究CMTM7在舌鳞状细胞癌(TSCC)组织中的表达及其对舌癌细胞SCC-15增殖和迁移能力的影响。方法用苏木精-伊红染色观察舌癌及癌旁组织的病理学形态,用免疫组化法检测CMTM7在舌癌及癌旁组织中的表达和分布,用实时荧光定量聚合酶链反应检测CMTM7在30对舌癌及癌旁组织中的表达情况。体外培养舌癌细胞系SCC-15,通过慢病毒构建CMTM7稳定过表达的细胞系,用EdU实验、平板克隆实验及Transwell实验检测SCC-15细胞的增殖及迁移能力。结果舌癌组织中癌细胞呈现浸润性分布,CMTM7在舌癌细胞中呈现均一的阳性表达,其表达量高于癌旁组织;30例患者的CMTM7 mRNA在舌癌组织中的表达水平较癌旁组织上调,其RNA绝对值为(3.44±0.65)×10-3,高于癌旁组织的RNA绝对值(1.18±0.21)×10-3,差异有统计学意义(P<0.01)。过表达CMTM7的舌癌细胞与对照组的EdU阳性细胞数分别为(51.94±7.96)和(16.92±3.89)个,迁移细胞数分别为(168.41±19.51)和(40.34±11.07)个,差异均有统计学意义(均P<0.05)。结论 CMTM7在舌癌组织中的表达高于癌旁组织,其促进舌癌发病进程的作用可能与增强舌癌细胞的增殖和迁移能力相关。 Objective To investigate the expression profile of CMTM7 in tongue squamous cell carcinoma(TSCC)and the effect on proliferation and migration of TSCC cells.Methods The distribution and expression of CMTM7 were detected by hematoxylin-eosinstaining and immunohistochemistry.CMTM7 expression was examined in 30 pairs of TSCC and paracancerous tissues by quantitative reverse transcription-polymerase chain reaction.In vitro culture of tongue cancer cell line SCC-15,stable overexpression of CMTM7 cell lines were constructed by lentivirus infection.Proliferation ability was explored by EdU test and colony assay.Migration ability was investigated through Transwell assay.Results The cancer cells showed infiltrative distribution in tongue carcinoma,and CMTM7 was uniformly expressed in tongue cancer cells,and its expression was higher than that in paracancerous tissues.The mRNA expression levels of CMTM7 in TSCC tissues and paracancer tissues were(3.44±0.65)×10-3 and(1.18±0.21)×10-3 with significant difference(P<0.01).The EdU positive cells in CMTM7-overexpressed cells and control cells were 51.94±7.96 and 16.92±3.89,and the migrated cells were168.41±19.51 and 40.34±11.07,the differences were statistically significant(all P<0.05).Conclusion The expression of CMTM7 in TSCC is higher than that in paracancerous tissues.Its role in promoting the pathogenesis of tongue cancer maybe related to the enhancement of proliferation and migration of TSCC cells.
