摘要 目的观察乳腺癌组织中促红细胞生成素产生肝细胞受体A2(EphA2)、肝配蛋白A1(EphrinA1)表达。方法采用免疫组织化学(IHC)法检测乳腺癌组织(选取2013年1月至2018年12月于大理大学第一附属医院行乳腺癌根治术或改良根治术且术前未经放化疗或其他治疗的女性患者术后标本250例)中EphA2、EphrinA1蛋白表达,用原位杂交(ISH)检测EphA2、EphrinA1 mRNA表达,间接免疫荧光法(IFA)检测EphA2、EphrinA1表达。组间采用χ2检验进行样本率比较、相关分析。结果250例乳腺癌病理标本中EphA2、EphrinA1蛋白阳性率分为74.80%、71.60%;EphA2、EphrinA1 mRNA阳性率分为83.60%、72.80%。EphA2、EphrinA1蛋白及相应mRNA的阳性率与临床分期、淋巴结转移、组织学分级明显相关(χ2=16.303、23.296、9.119、21.462、6.998、11.807、20.648、11.709、19.816、7.256、7.276、11.510,P值均<0.05)。IFA检测正常乳腺细胞中EphA2、EphrinA1几乎不表达而乳腺癌细胞中高表达。结论EphA2、EphrinA1可能与乳腺癌发生进展有关。 Objective To investigate the expression of erythropoietin-producing hepatomocellular A2(EphA2)and erythropoietin-producing hepatomocellular receptor interacting protein A1(EphrinA1)in breast cancer tissues.Methods The expression of EphA2 and EphrinA1 proteins in breast cancer tissues(From January 2013 to December 2018,a total of 250 postoperative pathological specimens were collected from female breast cancer patients at the First Affiliated Hospital of Dali University who had undergone radical mastectomies or modified radical mastectomies and had not been subjected to radiotherapy,chemotherapy,or any other special treatments before surgery)was detected by immunohistochemistry(IHC),the expression of EphA2 and EphrinA1 mRNA was detected by in situ hybridization(ISH),and the expression of EphA2 and EphrinA1 mRNA was detected by indirect immunofluorescence assay(IFA)after breast cancer cells were cultured.statistical programfor social sciences(SPSS)software was used to process the data,and the chi-square test was used for sample rate comparison and correlation analysis.Differences were statistically significant at P<0.05.Results The positive expression rates of EphA2 and EphrinA1 in 250 cases of breast cancer were 74.80% and 71.60% respectively.The positive expression rates of EphA2 mRNA and EphrinA1 mRNA were 83.60% and 72.80% respectively.The positive rates of EphA2 and EphrinA1 proteins and their corresponding mRNA in histopathological specimens of breast cancer were correlated with clinical stage,lymph node metastasis and histological grade(χ2=16.303,23.296,9.119,21.462,6.998,11.807,20.648,11.709,19.816,7.256,7.276,11.510,allP<0.05).IFA showed that EphA2 and EphrinA1 were hardly expressed in normal breast cells,while highly expressed in breast cancer cells.Conclusion EphA2 and EphrinA1 were highly expressed in breast cancer tissues and expected to be new targets for prognosis and treatment of breast cancer.
