乳腺癌组织中Yes相关蛋白1和真核翻译起始因子5A2的表达及其临床意义

摘要 目的探讨Yes相关蛋白1(YAP1)和真核翻译起始因子5A2(EIF5A2)在乳腺癌组织中的表达情况及其与乳腺癌临床病理指标的关系。方法收集2015年3月至2020年8月烟台山医院病理学确诊的77例乳腺癌组织和癌旁组织,使用免疫组织化学方法检测组织中YAP1蛋白和EIF5A2蛋白表达情况,结合临床资料行χ2检验分析。结果乳腺癌组织中YAP1蛋白阳性表达率为42.86%(33/77),明显低于癌旁组织中阳性表达率79.22%(61/77),两者差异有统计学意义(χ2=21.407,P<0.01),乳腺癌组织中EIF5A2蛋白阳性表达率为77.92%(60/77),明显高于癌旁组织中阳性表达率11.69%(9/77),两者差异有统计学意义(χ2=68.296,P<0.01);YAP1表达水平与乳腺癌组织学分级、TNM分期、淋巴结转移呈明显相关(χ2=8.968、4.442、4.345,P<0.05),差异有统计学意义。EIF5A2表达水平与乳腺癌肿瘤大小、组织学分化程度、TNM分期、淋巴结转移呈明显相关(χ2=4.230、6.497、6.912、5.544,P<0.05),差异有统计学意义。结论YAP1蛋白在乳腺癌组织中低表达、EIF5A2蛋白在乳腺癌组织中高表达与乳腺癌的侵袭性和恶性程度密切相关。 Objective To explore the expression of Yes-associated protein 1(YAP1)and eukaryotic translation initiation factor 5A2(EIF5A2)in breast carcinoma tissues,and to analyze their correlation with clinicopathological characteristics.Methods Immunohistochemistry was used to examine the expression levels of YAP1 protein and EIF5A2 protein in 77 cases of human breast carcinoma tissues and non-cancerous adjacent breast tissues(5 cm distance away from the margin of breast carcinoma)of patients pathologically confirmed from March 2015 to August 2020 on Yantai Mountain Hospital.The statistical analysis of chi-square test in combination with clinical data was carried out.Results The positive rate of YAP1 protein expression in breast carcinoma tissues[42.86%(33/77)]was significantly lower than that in non-cancerous adjacent breast tissues[79.22%(61/77),χ2=21.407,P<0.01].The positive rate of EIF5A2 protein expression in breast carcinoma tissues[77.92%(60/77)]was significantly higher than that in non-cancerous adjacent breast tissues[11.69%(9/77),χ2=68.296,P<0.01].The expression of YAP1 was related breast carcinoma to histology grade,tumor node metastasis stage(TNM stage)and lymph node metastasis(respectively,χ2=8.968,4.442,4.345,P<0.05).The expression of EIF5A2 was related breast carcinoma to tumor size,histology grade,TNM stage and lymph node metastasis(respectively,χ2=4.230,6.497,6.912,5.544,P<0.05).Conclusion Low expression of YAP1 and high expression of EIF5A2 in breast carcinoma tissues may be closely correlated with the invasion and malignant capacity of breast carcinoma.
