血府逐瘀汤对抑郁模型小鼠行为学及脑内单胺类神经递质的影响

摘要 目的:观察血府逐瘀汤对慢性不可预知性温和应激(CUMS)抑郁模型小鼠行为学及脑内单胺类神经递质水平变化的影响,探讨血府逐瘀汤的抗抑郁作用及其机制。方法:40只雄性CUMS抑郁模型小鼠随机分为正常对照组、模型组、血府逐瘀汤组和盐酸氟西汀组。除正常对照组外,其余3组小鼠均给予CUMS刺激,持续刺激3周制备抑郁模型。模型制备成功后,正常对照组、模型组使用生理盐水灌胃;血府逐瘀汤组小鼠按10.14 g/kg(含生药1 g/mL)剂量灌胃血府逐瘀汤;盐酸氟西汀组小鼠按剂量10 mg/kg(0.1%的盐酸氟西汀给药体积为10.14 mL/kg)灌胃盐酸氟西汀,每天2次,各组小鼠连续灌胃给药14 d,于末次给药后测定各组小鼠自主活动次数、悬尾与强迫游泳不动时间;并取脑组织测定单胺类神经递质,检测去甲肾上腺素(NE)、5-羟色胺(5-HT)、多巴胺(DA)水平变化。结果:与正常对照组比较,模型组小鼠水平运动次数、垂直运动次数均显著降低;悬尾不动时间、强迫游泳不动时间显著增加;小鼠5-HT、NE、DA含量显著降低(P<0.01,P<0.05)。与模型组比较,血府逐瘀杨组小鼠水平运动次数、垂直运动次数均显著增加;悬尾与强迫游泳不动时间显著降低;脑组织中NE、5-HT、DA水平明显升高(P<0.01,P<0.05)。结论:血府逐瘀汤具有明显的抗抑郁作用,其机制与上调脑内单胺类神经递质水平有关。 Objective:To study the effects of Xuefu Zhuyu decoction on the behavior and monoamine neurotransmitters of chronic mild unpredictable stress(CUMS)depression mice model and investigate its anti-depression effect and mechanism.Methods:Forty Kunming mice were randomly divided into normal control group,depression model group,Xuefu Zhuyu decoction group and fluoxetine hydrochloride group.Execpt the normal control group,the other three groups of rats were given CUMS to stimulate the production of CUMS depression model,then gavaged therapeutic drugs respectively for 14 d.Mice in each group were measured the number of independent activities,the time of tail suspension and forced swimming.All the mice were killed to get the brain to test the levels of NE,5-HT,DA by ELISA.Results:Compared with the normal group,the time of horizontal exercises and vertical exercises in the model group were reduced;the immobility time of tail suspension and the time of forced swimming immobility were increased;the content of 5-HT,NE,and DA in mice were decreased(P<0.01,P<0.05).Compared with the depression model group,the time of horizontal exercises and vertical exercises were increased in the Xuefu Zhuyu Yang group;and the time of tail suspension and forced swimming were reduced in Xuefu Zhuyu decoction group,and the contents of NE,5-HT,DA were increased in Xuefu Zhuyu decoction group(P<0.01,P<0.05).Conclusion:Xuefu Zhuyu decoction has a better anti-depressant effect whose mechanism is related to increasing the level of monoamine neurotransmitters in the brain.
